Abstract: To explore the effects of pressure, temperature, precooling and use of plastic bags on the preservation effect of mixed fresh-cut fruits and vegetables. In this Paper, apples, strawberries and potatoes are taken as the research objects. After fresh cutting, an appropriate amount of them is mixed, and the near-freezing point is set as -1°C. Seven control groups are set up: group A (normal pressure, normal temperature); Group B (atmospheric pressure, 4°C); Group C (10 kPa, -1°C); Group D (vacuum precooling, 10 kPa, -1°C); Group E (10 kPa, 4°C); Group F (vacuum precooling, 10 kPa, 4°C); Group G (without fresh bag, normal pressure, normal temperature) studied its preservation effect under different storage conditions within 9 days. Through the analysis and comprehensive evaluation of various indicators, the suitable storage and preservation methods of mixed fresh-cut fruits and vegetables are obtained. The results show that the preservation effect is more obvious at the later stage of the storage experiment, the final sensory score is the highest, the weight loss rate is the lowest, and the weight loss rate is reduced by 4.91% compared with the low pressure environment at 4°C. The use of plastic bags have the greatest impact on the weight loss rate and appearance of fruits and vegetables. Using plastic bags can reduce the weight loss by 48.48% on average compared with the same period, and the sensory score increases by 1 to 3 points. In the low pressure and high humidity environment, the weight of apple increases, and the final weight increases by 0.46% to 1.55%. The weight loss rate of strawberry is the highest, which can reach 22.54%. The vacuum pre-cooling process can reduce the weight of fruits and vegetables by 0.76% to 1.15%. Abstract: Objective: To improve the counting efficiency and realize the traceability of raw data. The applicability, accuracy, precision and repeatability of the automatic colony counter were verified by experiments, Determine the applicability of colony count in the detection of dairy products and raw and auxiliary materials. Methods: The plate pouring method and test piece method are selected for verification test, including psychrophilic bacteria, aerobic spores, thermophilic aerobic spores, lactic acid bacteria count, total number of colonies, coliform, mold, yeast, Staphylococcus aureus and other items. The manual count is compared with the automatic colony counter to determine its feasibility, applicability and accuracy. Results: The colony count of normal Petri dish without background interference was compared with that of 3M test piece. The paired t-test was carried out by visual method and instrument method, P = 0.17, greater than 0.05. There was no significant difference in statistical results, and the colored medium had no significant effect on automatic counting. The counting effect of the instrument is poor when there are needle shaped colonies, the color of colonies is close to that of the culture medium, the contour of colonies is not obvious, the colonies spread greatly and the edges of colonies are irregular, there are food particles and colonies in the culture medium at the same time, or the color of colonies is close to that of food particles, which can not meet the accuracy requirements. Conclusion: For samples with uniform texture, the accuracy and precision of automatic colony counting are relatively good, the work efficiency is high, the operation time of experimental personnel is saved, and the original experimental data and images can be stored in the instrument for a long time. It is a counting method worthy of popularization in the laboratory. Take milk powder as an example, the uncertainty of graphite furnace atomic absorption spectrometry method 1 in GB 5009.12-2017 National Food Aafety Standard Determination of Lead in Foods was evaluated by referring to JJF 1059.1-2012 Evaluation and Expression of Measurement Uncertainty. When the standard result of lead content in milk was 0.095 mg/kg, the synthetic standard uncertainty of the determination result was 0.0022 mg/kg, k=2, and the extended uncertainty was 0.005 mg/kg, and the result could be expressed as (0.095±0.005) mg/kg. The uncertainty of measurement results is mainly due to the uncertainty component introduced by test repeatability and standard solution concentration.
