Abstract: Somatic embryogenesis (SE) is an important type of plant regeneration, which can be used as a recipient system of genome editing. It is vital to optimize the SE system to improve the mutated somatic cells regenerated into fertile plants after transformation. 2,4-D was used as inducer of somatic embryogenesis from mature seeds. Arabidopsis wild type Col-0 and ABA biosynthesis double mutant nced6 / 9 were grown in 15°C and 25°C respectively, to investigate the effect of sampled plants growth conditions on the SE rate of their seeds and to optimize SE protocol. On the other hand, construct Pbi121-U6-26-Cas9-sgRNA-ELF6-FastR was used for genome editing. The positive seeds after floral dip were separated into two parts: some for sowing and the rest seeds went though SE induction. The efficiency of gene editing was compared by sequencing. Our results show that the SE rate of nced6 / 9 seeds from 25°C was significantly lower than that from 15°C. When transgenic positive seeds were sown directly, the target sequence was not found to be edited; However, when seeds went though somatic embryogenesis protocol, gene editing were found around PAM sequence, suggesting that SE system could improve gene editing after transformation. The sampled plants grown at low temperature (15°C) may affect SE ability of their seeds by ABA level. Abstract: Objective: During pregnancy, pregnant women are prone to abnormal liver function due to excessive ideological burden and other reasons, which seriously endangers the physical health of pregnant women and the normal development of the fetus. The therapeutic effect of psychological nursing in many diseases has received more and more attention, but its clinical value in pregnant patients with non-viral abnormal liver function is still unclear. Therefore, this project studied the clinical value of psychological nursing in pregnant patients with non-viral abnormal liver function. Methods: 70 pregnant patients with non-viral abnormal liver function in our hospital from April 2020 to April 2022 were recruited and randomly divided into control group (35 cases) and observation group (35 cases). The control group received regular pregnancy tests combined with routine intervention, and the observation group received psychological nursing on the basis of the control group. One month later, the liver function and psychological changes of the two groups were compared. Results: The indexes of liver function (alanine aminotransferase [ALT] and aspartate aminotransferase [AST]), self-rating anxiety scale (SAS) score, and self-rating depression scale (SDS) in the observation group were lower than those in the control group (P< 0.05). While the comprehensive assessment of quality of life (GQOLI-74) was significantly higher than that of the control group (P<0.05). Abstract: The genus Roscoea is an ideal taxa for evaluating speciation patterns in global biodiversity hotspots. It is related to the genus Cautleya based on the phylogenetic relationship, and they were divided by the early rapid uplift of Himalayan-Tibet Plateau and the associated climate change, in the late Eocene to the early Oligocene (~30-40 Ma). Roscoea originated in Assam, India and both was separated into two clades in the Oligocene/Miocene boundary (about 22-25 Ma), while two lineages were formed and have been evolving independently (about 13-38 Ma). Abstract: [Objective] By exploring the biological characteristics and artificial large-scale breeding technology of Papilio polytes, the artificial large-scale breeding technology system of P. polytes was established in Chongqing, which provided a strong guarantee for the resource development, utilization and protection of P. polytes. [Methods] Through the method of field introduction and artificial breeding of P. polytes, under the conditions of 22-30°C and 60%~75% humidity, P. polytes was bred in the Insect Innovation base of Chongqing Normal University. The detailed growth and biological characteristics of P. polytes in each developmental period were observed and recorded, and the technical method system of artificial large-scale breeding of P. polytes in Chongqing was summarized. [Results] The development process and biological characteristics of each developmental stage of P. polytes were initially mastered. P. polytes was successfully bred, and a complete set of artificial large-scale breeding technology system was established. [Conclusion] Using this technique, the growth and development of each stage of P. polyte was good, and the offspring could be successfully bred.
